Rubella haemagglutination inhibition tests

The haemagglutination inhibition test detects antibody classes IgG and IgM. The test is based on the ability of the rubella antigen to agglutinate chick erythrocytes = haemagglutination. The agglutination is inhibited by specific rubella antibodies in the patient serum = haemagglutination inhibition (HI). Non-specific inhibition factors in the serum must be removed through an adsorption procedure.

Haemolysis-in-gel test for rubella

The haemolysis-in-gel test for rubella is particularly suitable for detecting low or borderline titres in the HIT test. Through the use of chick erythrocytes in the test, non-specific reactions are rare in pathological sera (5%); they can be detected using the control plates. The test can also contribute to diagnostic clarification of a fresh infection: IgM antibodies, which arise on the 1st to 2nd day after the onset of illness, are detected by the HIT but not by the HiG. IgG antibodies are detected starting on the 2nd day in the HIT and on the 5th to 7th day in the HiG. A difference in titre between the two methods indicates an acute infection and requires further serological examinations.
